We develop enable technologies for making synthetic DNA using parallel and miniaturized platform. We are interested in methods for making DNAs (and thus RNAs) as basic elelments/constructs for functional DNA/RNA clusters and proteins/protein comlexes, molecular devices of designed structures and functions, and synthetic biosystems. Genomes are extremely complex molecular machines, which contain essential information governing our lives. We have developed a novel solution photochemistry so that the regular nucleic acid chemistry are carried out for making DNA CHIPs that is used to read DNA sequences, probe DNA structures, and discover base variations/mutations. In a massively parallel fashion, these experiments are performed in parallel in a miniaturized setting. This is a new format of reaction and assay, involving high throughput combinatorial synthesis, biological sample processing, and bioinformatic computing. Our research projects include BIOCHIP-based DNA synthesis and synthetic DNA applications in RNA and protein synthesis, design and develop enzymatic systems for biosynthesis and metabolic reactions.

 

we are also interested in the metabolic engineering of thermo-tolerant microbes for the fast growth under elevatated temperature. Now we are focusing on the xylose assimilation of Yeast of K. marixius. The engineered K. marixainus constructed in our lab could produced xylitol and ethanol with very high productivity already. Further work will try to improve the tolerance of inhibitor in lignocellusic hydrosates.
